Matino is located on the last slopes of the Salento Hills at 75 m above sea level, 10 km from the Ionian coast and 25 km from the extreme tip of the Salento peninsula represented by Capo di Leuca. The historic center extends on the hill called S. Ermete, nestled between two prehistoric gullies (karstic ravines) respectively named Universo on the north side and Reale on the south side. The territory, strongly karstic, is devoid of surface water courses but very rich in underground water courses fed by the so-called Vore, natural sinkholes of rainwater located in natural depressions in the ground. The municipality, which extends over an area of 26.28 km², reaches a minimum height of 38 m above sea level and a maximum height of 172 m above sea level. The town dominates the valley called Taviano-Matino, which represents one of the most fertile areas of Salento. The predominant crops are olive and vine, but with large spaces dedicated to arable land and greenhouses, especially flowers.
